چکیده

An increasing number of investigators (2-5) have postulated the occurrence of an exchange reaction catalyzed by transaldolase between glyceraldehyde S-phosphate and fructose 6phosphate to explain the unequal labeling of the 6 carbons of hexoses which is observed in tracer experiments. Transaldolase accepts the dihydroxyacetone moiety from sedoheptulose 7-phosphate or fructose B-phosphate to form a dihydroxyacetone-enzyme intermediate (6). In the presence of an acceptor aldehyde such as glyceraldehyde 3-phosphate or erythrose 4-phosphate the dihydroxyacetone moiety is transferred to form the corresponding keto sugar-phosphate as follows:

Sources of hepatic glycogen synthesis during an oral glucose tolerance test: Effect of transaldolase exchange on flux estimates.

Sources of hepatic glycogen synthesis during an oral glucose tolerance test were evaluated in six healthy subjects by enrichment of a 75-g glucose load with 6.67% [U-(13)C]glucose and 3.33% [U-(2)H(7)]glucose and analysis of plasma glucose and hepatic uridine diphosphate-glucose enrichments (sampled as urinary menthol glucuronide) by (2)H and (13)C nuclear magnetic resonance. The direct pathway...

A coupled reaction catalyzed by the enzymes transketolase and transaldolase.

The formation of pentose phosphate from hexose monophosphate involves either the oxidative pathway from glucose 6phosphate through 6-phosphogluconate or the series of nonoxidative transfer reactions catalyzed by transketolase and transaldolase, in which sedoheptulose 7-phosphate is an intermediate (1). In mammalian tissues the nonoxidative pathway appears to play a major role (2-5), accounting ...

Lactose synthesis. VI. Labeling of lactose precursors by glycerol-1,3-C-14 and glucose-2-C-14.

It is generally agreed that blood glucose is the major source of carbon for the synthesis of both hexose portions of lactose by the mammary gland (1, 2), but the exact mechanism of lactose synthesis remains in doubt. An abridged outline of some possible pathways of labeled carbon in the biosynthesis of lactose is presented in Fig. 1. In Reaction A of Fig. 1, uridine diphosphate galactose is the...

Initial Reactions in the Metabolism of D- and L-Glyceraldehyde by Rat Liver* BERNARD R. LANDAut AND WILFRIED MERLEVEDE WITH THE TECHNICAL ASSISTANCE OF HOLLIS

There appear to be several pathways in liver by which Dand n-glyceraldehyde can be converted to dihydroxyacetone phosphate and n-glyceraldehyde phosphate. Thus, the following reactions have been described in liver preparations: phosphorylation of n-glyceraldehyde to n-glyceraldehyde phosphate (Fig. 1; Pathway A) (2); formation of n-glyceraldehyde phosphate from n-glyceraldehyde via n-glyceric a...

Replacement of a phenylalanine by a tyrosine in the active site confers fructose-6-phosphate aldolase activity to the transaldolase of Escherichia coli and human origin.

Based on a structure-assisted sequence alignment we designed 11 focused libraries at residues in the active site of transaldolase B from Escherichia coli and screened them for their ability to synthesize fructose 6-phosphate from dihydroxyacetone and glyceraldehyde 3-phosphate using a newly developed color assay. We found one positive variant exhibiting a replacement of Phe(178) to Tyr. This mu...
